They have to visit Chorley, a club playing in Vanarama North, three levels above the Hillmen in the non-league pyramid.
Club chairman Dave Atkinson is the first to admit that the odds are against his side. Even so, he's hoping Glossop might benefit from some of that legendary FA Cup magic.
"Chorley are big favourites and we know it's going to be very tough, but they've been to watch us so at least we know they're not taking us lightly," he commented.
"Every round of the cup throws up a shock result and maybe this time it'll be Glossop who spring the surprise."
The Hillmen go into the tie on the back of an 8-0 victory at Stockport Sports in which Jason Carey, signed from Runcorn Town over the summer, scored a hat-trick.
Chorley, who are managed by Manchester City and Blackburn midfielder Gary Flitcroft, are reigning Evo-stik NPL champions and third in Vanarama North, only two points behind leaders Fylde who were promoted form the NPL premier division through the play-offs.
The Lancashire side's top scorer is former kick-boxing champion James Dean who has played for Hyde and Stalybridge.
Glossop have won through all three rounds of the FA Cup so far, picking up almost £6,500 along the way. Even so, Atkinson remains annoyed that there have been midweeks when his side have not played, allowing Runcorn Linnets to steal a march on them at the top of the NWCL premier division table.
He said; "I know I seem to talk about this every year but the Linnets have played three more league games than we have. I would never think of moaning about the FA Cup but there's something wrong when you think they're getting league games and we aren't. In the new year they'll be asking us to play three matches a week."
